Abstract

Dairy cattle farms in Sonora are characterized by continuous annual production, which is seriously affected by adverse environmental temperature and humidity conditions that prevail most of the year, as they exceed the cow's thermal comfort threshold, leading to heat stress. Despite the challenge these conditions represent for milk production systems, it is possible to observe cows with the ability to maintain efficient production parameters, which is attributed to a genetic basis. Therefore, the objective of this study was to evaluate a polymorphism of the TLR4 gene as a genetic marker for traits associated with milk production in Holstein cows managed under heat stress. The study included 264 lactating cows from two farms located in the Yaqui Valley. Total milk production during lactation was recorded and adjusted to 305 days. Climatological data were obtained to calculate the THI, which indicated the presence of heat stress from April to October. Blood samples were obtained from each cow and processed for DNA extraction, which was used for genotyping of a single nucleotide polymorphism (SNP) of the toll-like receptor 4 (TLR4) gene by TaqMan assay and qPCR test. A mixed effects statistical model validated the predictive effect (P < 0.05) of the SNP rs8193046 on four traits associated with milk production and confirmed the additive behavior of the TLR4 gene. The favorable genotype was the TT genotype, where each T allele showed a contribution (P < 0.01) in the improvement of the four parameters evaluated. In conclusion, the SNP rs8193046 of the TLR4 gene is proposed as a genetic marker associated with milk production in Holstein cows managed under heat stress.
